how do I hook the tube with water line to water inlet valve in the icemaker MFI2569VEB2 Maytag Refrigerator

Thenak you for your question.  I believe from the description and problem you are describing a "John Guest" fitting.  On the outer edge of the connector is a small ring that can be pushed with a fingernail or small screwdriver to release the hose.  When the hose is inserted it will snap into place.  The following link to a video may be helpful: http://www.youtube.com/watch?v=-Avzf-jsHyM .  I hope this helps.

The only thing about the kit is that it is stated that if the ice maker is not turned off then the freezing in the refrigerator section might happen.

Is the ice maker turned off or is it still just not working. If not working then add some water to the mold if it is empty and see if that water is frozen and harvested.

There wash an known issue with cold air leaking from the ice maker compartment causing the refrigerator section to freeze. The kit will help this problem.

I'd suggest first trying to correct the ice maker problem. You can reply with any additional details for further assistance.
