Does your video card support multiple monitors? You may need to update your drivers. Is this on Windows? If so, right click on your Desktop (away from any icons) and choose Properties. Then look for the appropriate settings.

If that doesn't help, I'll need to know what version of Windows you are using (XP? Vista? 7) and exactly what you tried.

You may also need to turn the computer off (not just reboot, shutdown completely) and then turn it back on for it to realize there is new hardware attached.
